  • 小鼠腎炎模型的制備及雙氫青蒿素對其炎癥因子釋放的影響

    【摘要】 目的 制備小鼠腎炎模型并觀察雙氫青蒿素(dihydroartemisinine,DHA)對模型小鼠細(xì)胞因子腫瘤壞死因子-α(tunor necrosis factor,TNF-α)和白細(xì)胞介素-6(inter leukin-6,IL-6)的影響以及小鼠腎臟的病理變化?!》椒ā∪⌒坌岳ッ鞣N小鼠120只, 隨機分為正常對照組、脂多糖(lipopolysaccharides,LPS)組、LPS+腎勻漿組及DHA治療組;分別于12、24、48 h取血,酶聯(lián)免疫吸附試驗檢測血清中TNF-α和IL-6的含量,蘇木精-伊紅染色法觀察小鼠腎臟的病理變化?!〗Y(jié)果 造模48 h LPS+腎勻漿組小鼠腎小球出現(xiàn)炎性細(xì)胞浸潤,而正常對照組未見異常;LPS組及 DHA治療組僅有輕微的病理改變。LPS刺激使小鼠血清TNF-α和IL-6含量高于正常水平(Plt;0.01),但有隨時間不斷下降的趨勢;LPS+腎勻漿組較正常對照組TNF-α和IL-6含量升高(Plt;0.01);DHA可顯著下調(diào)模型小鼠血清TNF-α的水平(Plt;0.01),但對IL-6的影響相對較?。≒gt;0.05)?!〗Y(jié)論 運用改良的造模方法LPS+腎勻漿建立腎炎模型效果良好;DHA可以調(diào)節(jié)模型小鼠炎癥因子TNF-α和IL-6的釋放,具有一定的改善模型小鼠腎炎癥狀的作用?!続bstract】 Objective To establish mice nephritis models, detect the serum level changes of cytokines tumor necrosis factor α (TNF-α) and interleukin-6 (IL-6) in the model mice treated by Dihydroartemisinin (DHA), and observe the physiological changes of the mice kidneys. Methods One hundred and twenty male Kunming mice were randomly divided into 4 groups: control group, lipopolysaccharides (LPS) group, LPS plus kidney homogenate group, and DHA treated group. The level of cytokines TNF-α and IL-6 in the serum were detected by enzyme-linked immunoabsordent assey at hour 12, 24, and 48, respectively. Pathological changes were observed by hematoxylin: eosin staining. Results At the time hour 48 after the establishment of the model, inflammatory cell infiltration was observed in the glomerulus of the LPS plus kidney homogenate group, but no abnormality was found in the control group. There were only slight pathological changes in mice models of the LPS group and the DHA treated group. The serum level of TNF-α and IL-6 increased remarkably after the treatment of LPS (Plt;0.01), but declined as time went by. The level of TNF-α and IL-6 increased significantly in LPS plus kidney homogenate group compared with the control group (Plt;0.01). DHA could significantly decrease the TNF-α level in the serum (Plt;0.01), but had a low influence on IL-6 (Pgt;0.05). Conclusion The modified LPS plus kidney homogenate has a good result in model establishing. DHA can regulate the release of TNF-α and IL-6 in the model mice, and may have certain good effects on ameliorating the nephritis pathological changes.

  • 天然藥物青蒿素及其衍生物的作用機理研究進(jìn)展

    目的 探討天然藥物青蒿素及其衍生物的藥理作用及機理。方法 復(fù)習(xí)近年來國內(nèi)、外相關(guān)文獻(xiàn)并進(jìn)行綜述。結(jié)果 青蒿素及其衍生物結(jié)構(gòu)獨特、高效低毒,除具有抗瘧疾作用外,亦被證明具有抗腫瘤、調(diào)節(jié)免疫功能、抗菌、抗孕、抗纖維化、清熱解毒等藥理作用。結(jié)論 青蒿素及其衍生物通過不同的途徑發(fā)揮藥理作用,具有較好的應(yīng)用前景。

  • 青蒿素誘導(dǎo)急性胰腺炎胰腺腺泡細(xì)胞凋亡的實驗研究

    目的 探討青蒿素(artemisinin)對急性胰腺炎胰腺腺泡細(xì)胞凋亡的影響,并了解是否減輕急性胰腺炎嚴(yán)重程度。方法 腹腔內(nèi)注射雨蛙素(cerulein)誘導(dǎo)大鼠胰腺炎模型建立,設(shè)正常對照組、胰腺炎組及青蒿素組,通過原位末端標(biāo)記(TUNEL)法檢測各組胰腺組織中腺泡細(xì)胞的凋亡指數(shù),并用分光光度法檢測髓過氧化物酶(MPO)活性。采用兩步酶消化法分離正常大鼠胰腺腺泡細(xì)胞,離體分組培養(yǎng),采用丫碇橙(AO)染色及caspase3活性的檢測,評估青蒿素對胰腺腺泡細(xì)胞凋亡的影響,并檢測各組培養(yǎng)液中的乳酸脫氫酶(LDH)及淀粉酶(AMS)活性,評估胰腺腺泡細(xì)胞酶的釋放。結(jié)果 在體實驗中,青蒿素組與胰腺炎組比較,胰腺腺泡細(xì)胞凋亡指數(shù)明顯增高(P<0.05),MPO活性明顯減低(P<0.05)。體外實驗中,青蒿素組胰腺腺泡細(xì)胞凋亡指數(shù)及caspase3活性較胰腺炎組明顯增高(P<0.05),培養(yǎng)液中的LDH和AMS活性也明顯降低(P<0.05)。結(jié)論 青蒿素可促進(jìn)大鼠急性胰腺炎胰腺腺泡細(xì)胞凋亡,減少胰酶釋放及中性粒細(xì)胞激活,可減輕急性胰腺炎嚴(yán)重程度。
