目的  探討青蒿素(artemisinin)對急性胰腺炎胰腺腺泡細(xì)胞凋亡的影響,并了解是否減輕急性胰腺炎嚴(yán)重程度。
方法  腹腔內(nèi)注射雨蛙素(cerulein)誘導(dǎo)大鼠胰腺炎模型建立,設(shè)正常對照組、胰腺炎組及青蒿素組,通過原位末端標(biāo)記(TUNEL)法檢測各組胰腺組織中腺泡細(xì)胞的凋亡指數(shù),并用分光光度法檢測髓過氧化物酶(MPO)活性。采用兩步酶消化法分離正常大鼠胰腺腺泡細(xì)胞,離體分組培養(yǎng),采用丫碇橙(AO)染色及caspase3活性的檢測,評估青蒿素對胰腺腺泡細(xì)胞凋亡的影響,并檢測各組培養(yǎng)液中的乳酸脫氫酶(LDH)及淀粉酶(AMS)活性,評估胰腺腺泡細(xì)胞酶的釋放。
結(jié)果  在體實驗中,青蒿素組與胰腺炎組比較,胰腺腺泡細(xì)胞凋亡指數(shù)明顯增高(P<0.05),MPO活性明顯減低(P<0.05)。體外實驗中,青蒿素組胰腺腺泡細(xì)胞凋亡指數(shù)及caspase3活性較胰腺炎組明顯增高(P<0.05),培養(yǎng)液中的LDH和AMS活性也明顯降低(P<0.05)。
結(jié)論  青蒿素可促進大鼠急性胰腺炎胰腺腺泡細(xì)胞凋亡,減少胰酶釋放及中性粒細(xì)胞激活,可減輕急性胰腺炎嚴(yán)重程度。
