【摘要】目的 探討保存溫度對移植胰腺功能的影響,確定大鼠胰腺的最適保存溫度。
方法 采用原位灌注、單純低溫保存及建立同系大鼠異位全胰十二指腸移植模型,按不同保存溫度(0 ℃、4 ℃、8 ℃、12 ℃)進行分組,保存6 h后進行移植,通過光鏡觀察胰腺組織結構變化,高壓液相色譜法檢測保存6 h后移植物ATP和總腺苷核苷酸(TAN)含量。移植24 h后,檢測血糖、血清中脂肪酶及淀粉酶含量,測定移植胰腺組織中髓過氧化酶(MPO)活性,并進行組織學觀察。
結果 4 ℃組保存6 h后和移植24 h后的胰腺組織結構損傷明顯輕于其它組。保存6 h后胰腺組織中ATP和TAN水平在4 ℃組、0 ℃組、8 ℃組和12 ℃組依次降低,差異有顯著性意義(P<0.05)。移植24 h后血糖、血清脂肪酶及胰腺組織中MPO水平在4 ℃組、0 ℃組、8 ℃組和12 ℃組依次增高,差異均有顯著性意義(P<0.05)。結論4 ℃對大鼠胰腺是最適保存溫度,它能夠明顯減輕大鼠胰腺的冷損傷程度。
