【摘要】目的觀察誘導(dǎo)血紅素氧合酶-1(HO-1) 生成能否減輕老年供肝移植后缺血再灌注損傷。
方法取老年(16個月齡)SD大鼠5只和成年(3個月齡)大鼠5只,分別測定其肝臟組織中超氧化物歧化酶(SOD)和過氧化氫酶(CAT)的活性及維生素E(Vit E)、維生素C(Vit C)和丙二醛(MDA)含量; 另取老年SD大鼠60只作為供體,隨機均分為血晶素(Hemin)組和對照組,分別于取肝前24 h腹腔注射Hemin或生理鹽水,然后將其肝臟移植給同種大鼠,同時觀察再灌注后肝臟組織學(xué)變化和細胞凋亡。
結(jié)果老年大鼠肝臟組織中SOD活性明顯低于成年大鼠,Vit C含量亦少(P<0.05),而MDA明顯增高(P<0.05)。用Hemin預(yù)處理的老年SD大鼠供肝在切取前,SOD活性增加,Vit E、Vit C含量增加,MDA含量降低(P<0.05); 肝移植后血清ALT明顯降低,肝組織HO-1活性明顯增高,肝臟組織學(xué)檢查示再灌注后凋亡細胞明顯減少(P<0.05)。
結(jié)論老年SD大鼠肝臟存在氧化應(yīng)激和脂質(zhì)過氧化狀態(tài),HO-1可通過改善這種狀態(tài)而減輕其缺血-再灌注損傷。
