【摘要】目的探討細(xì)菌性腹膜炎致多器官功能不全綜合征(MODS)大鼠結(jié)腸運(yùn)動(dòng)的變化和細(xì)胞因子白介素6(IL6)、腫瘤壞死因子α(TNFα)及誘生性一氧化氮合成酶(iNOS)對(duì)結(jié)腸運(yùn)動(dòng)的影響。
方法將Wistar大鼠隨機(jī)分成正常對(duì)照組和MODS組; 分別觀測(cè)2組大鼠排便的糞點(diǎn)數(shù)、結(jié)腸肌條的振幅、結(jié)腸平滑肌細(xì)胞長(zhǎng)度以及血清一氧化氮(NO)的變化; 利用免疫組化和逆轉(zhuǎn)錄聚合酶鏈反應(yīng)(RTPCR)方法檢測(cè)結(jié)腸平滑肌IL6、TNFα和iNOS表達(dá)的變化。
結(jié)果MODS組排便的糞點(diǎn)數(shù)和肌條的振幅均明顯低于正常對(duì)照組(P<0.05); IL6、TNFα和iNOS蛋白表達(dá)在正常對(duì)照組為陰性表達(dá),在MODS組為陽性表達(dá); 在正常對(duì)照組未檢出IL6 mRNA、TNFα mRNA和iNOS mRNA的表達(dá),而在MODS組,IL6 mRNA、TNFα mRNA和iNOS mRNA均呈陽性表達(dá); MODS組平滑肌細(xì)胞的長(zhǎng)度和血清NO水平均較正常對(duì)照組明顯增高(P<0.01)。
結(jié)論MODS后結(jié)腸運(yùn)動(dòng)功能障礙與iNOS、IL6和TNFα密切相關(guān)。
