目的了解上皮型鈣黏附分子(E-cadherin)下調(diào)對(duì)腫瘤細(xì)胞浸潤(rùn)能力的影響。
方法選用人胰腺癌細(xì)胞株JHP-1,經(jīng)E-cadherin反義寡核苷酸(ASODN)作用,應(yīng)用免疫細(xì)胞化學(xué)染色、Western blot法檢測(cè)細(xì)胞E-cadherin蛋白表達(dá)情況,細(xì)胞浸潤(rùn)MTT比色法檢測(cè)該細(xì)胞對(duì)基底膜的浸潤(rùn)能力。
結(jié)果經(jīng)E-cadherin ASODN作用后, JHP-1細(xì)胞膜E-cadherin表達(dá)減弱, E-cadherin蛋白含量與對(duì)照組相比明顯降低; JHP-1細(xì)胞對(duì)基底膜浸潤(rùn)能力明顯高于對(duì)照組(P<0.001)。
結(jié)論E-cadherin與腫瘤細(xì)胞的浸潤(rùn)能力有關(guān)。
