目的  了解在體外條件下不同濃度的雌二醇對(duì)不同性質(zhì)的乳腺組織原代細(xì)胞增殖的影響。
方法  用膠原酶消化法獲取乳腺癌患者的各類乳腺組織原代細(xì)胞,并用MTT法檢測(cè)乳腺癌組織、癌旁組織和正常乳腺組織原代細(xì)胞在體外培養(yǎng)下,給予不同濃度雌二醇時(shí)細(xì)胞的生長(zhǎng)增殖情況。
結(jié)果  雌激素在體外對(duì)雌激素受體(ER)表達(dá)陽(yáng)性的乳腺癌組織及癌旁組織原代細(xì)胞有明顯的刺激生長(zhǎng)作用; 對(duì)正常乳腺組織細(xì)胞的促生長(zhǎng)作用相對(duì)較弱,而且與腫瘤組織的ER表達(dá)情況無關(guān)。
結(jié)論  當(dāng)體內(nèi)的雌二醇水平≥103 pmol/L時(shí),乳腺癌的發(fā)病風(fēng)險(xiǎn)和復(fù)發(fā)風(fēng)險(xiǎn)會(huì)顯著增加。
