目的:探討表沒食子兒茶素沒食子酸酯(EGCG)對(duì)乳腺癌細(xì)胞MCF7生長的影響及對(duì)乳腺癌細(xì)胞MDAMB231遷移的影響。方法:MCF7細(xì)胞培養(yǎng)貼壁之后,加入EGCG處理,2d后收集蛋白,采用Western Blot檢測磷酸化p38絲裂原活化蛋白激酶(phosphop38MAPK)的表達(dá);同樣處理后收集活細(xì)胞,用細(xì)胞計(jì)數(shù)法檢測細(xì)胞的存活;取對(duì)數(shù)生長期的MDAMB231細(xì)胞,分至6孔板培養(yǎng),使用EGCG處理后,采用細(xì)胞劃線法探測乳腺癌細(xì)胞的遷移。結(jié)果:使用EGCG處理乳腺癌細(xì)胞后,phosphop38MAPK的表達(dá)降低,EGCG處理乳腺癌細(xì)胞4d后其增殖率降低50%,遷移活性降低。結(jié)論:EGCG處理乳腺癌細(xì)胞能抑制腫瘤細(xì)胞的生長以及遷移,這與p38MAPK信號(hào)通路相關(guān)。
