目的:檢測李斯特菌DNA對小鼠H22肝癌Bcl2基因表達(dá)的影響,探討細(xì)菌DNA對腫瘤的抑制作用。方法:建立H22肝癌小鼠模型,從模型建立后的第一天開始,隨機(jī)分組,分別給予李斯特菌DNA液(實驗組)和生理鹽水(對照組)。從癌組織中提取總RNA,設(shè)計目的基因引物和內(nèi)參照GAPD引物。采用RT-PCR方法,分別檢測實驗組及對照組小鼠瘤體Bcl-2基因的表達(dá),應(yīng)用Bio-RAD凝膠成像系統(tǒng)及在Quantity One軟件輔助下分析測定表達(dá)結(jié)果。結(jié)果:實驗組小鼠瘤體Bcl-2表達(dá)低于對照組,經(jīng)統(tǒng)計學(xué)處理,兩者有顯著性差異。結(jié)論:李斯特菌DNA對H-22肝癌小鼠腫瘤有抑制作用。
