目的  探討自由基清除劑依達(dá)拉奉( ED) 對膿毒癥致ALI 的保護(hù)作用。方法  24 只雄性Wistar 大鼠隨機(jī)分成3 組: 對照組( NS組) 、模型組( LPS 組) 及依達(dá)拉奉治療組( ED 組) 。LPS 組和ED 組采用尾靜脈注射LPS( 10 mg/ kg) 建立ALI 模型, ED 組隨后立即尾靜脈注射依達(dá)拉奉( 3 mg/kg) 。LPS 注射6 h后留取肺標(biāo)本, 分別測定肺組織濕/干重比值( W/D) 和肺組織勻漿中髓過氧化物酶( MPO) 、丙二醛( MDA) 和超氧化物歧化酶( SOD) 含量, 觀察肺組織病理改變及肺組織核因子κB( NF-κB) 表達(dá)情況。結(jié)果  LPS 組肺組織MPO、MDA 含量、W/D、肺損傷評分及肺組織NF-κB 表達(dá)均高于NS組( P 均 lt;0. 01) , 肺組織SOD 含量低于NS 組( P  lt;0. 01) 。ED 組肺組織MPO、MDA 含量、W/D、肺損傷評分及肺組織NF-κB 表達(dá)均低于LPS組( P 均 lt;0. 01) , 仍高于NS 組( P 均 lt;0. 01) ;ED 組肺組織SOD 含量高于LPS 組( P  lt;0. 01) , 仍低于NS 組( P  lt;0. 01) 。結(jié)論  依達(dá)拉奉可以減輕LPS 誘導(dǎo)的大鼠ALI, 其機(jī)制可能與清除ROS, 降低了NF-кB 信號轉(zhuǎn)導(dǎo)通路的活化, 進(jìn)而減輕炎癥級聯(lián)放大效應(yīng)有關(guān)。
