為研究肝細胞低密度脂蛋白受體對膽汁中膽汁酸的影響及其在膽囊結(jié)石過程中可能的機理,采用高膽固醇膳食誘發(fā)兔膽囊結(jié)石模型,對進食后1、2、3、4周組及對照組動物,采用雙波長薄層掃描法測定膽汁中膽汁酸,125I-LDL放免標記法測定肝細胞低密度脂蛋白受體(LDLr)活性。結(jié)果: 高膽固醇膳食后2、3、4周組分別有4/10,6/10,7/10出現(xiàn)膽囊結(jié)石; 膽汁中甘氨膽酸明顯降低(P<0.05);125I-LDL與肝細胞低密度脂蛋白受體(LDLr)的最大結(jié)合力(Bmax)逐漸下降(P<0.05),解離常數(shù)(Kd)值逐漸升高(P<0.05)。上述結(jié)果提示: 肝細胞低密度脂蛋白受體活性下降,可能導(dǎo)致膽汁中甘氨膽酸減少,致成石性膽汁形成。
