目的 探討生長(zhǎng)抑素類似物SMS201995 (SMS) 對(duì)人膽管癌細(xì)胞體外增殖和凋亡的調(diào)控作用。方法采用人膽管癌細(xì)胞系SKChA1,應(yīng)用生長(zhǎng)曲線、流式細(xì)胞儀、片段DNA瓊脂糖凝膠電泳、Annexin VFITC技術(shù)及流式細(xì)胞儀間接免疫熒光技術(shù)等方法進(jìn)行檢測(cè)和觀察。結(jié)果在SMS 作用下,SKChA1細(xì)胞生長(zhǎng)受到抑制,短時(shí)使細(xì)胞阻滯于G0/G1期。SMS處理SKChA1細(xì)胞后, Annexin VFITC標(biāo)記的凋亡細(xì)胞增多, DNA凝膠電泳顯示典型的凋亡梯狀圖譜,同時(shí)SMS可使細(xì)胞bcl2蛋白表達(dá)下降,使bax蛋白表達(dá)升高。結(jié)論SMS 可明顯抑制SKChA1細(xì)胞生長(zhǎng),并能誘導(dǎo)該細(xì)胞凋亡。該細(xì)胞凋亡機(jī)理可能與凋亡調(diào)控基因bax和bcl2的表達(dá)有關(guān)。
