• 昆明醫(yī)學(xué)院第二附屬醫(yī)院肝膽胰一病區(qū)（昆明650101）;

目的  探討希羅達（Xeloda）對人肝癌組織AFP mRNA表達的影響。
方法  抽提所收集的希羅達治療組及對照組肝癌組織標(biāo)本總RNA（共41例），采用逆轉(zhuǎn)錄巢式聚合酶鏈反應(yīng)（RTnested PCR）技術(shù)檢測希羅達治療組及對照組AFP mRNA的表達情況。
結(jié)果  以RT-nested PCR擴增的AFP基因片段為101 bp，與原設(shè)計一致。在希羅達治療組中，21例肝癌組織標(biāo)本中有12例AFP mRNA呈陽性表達，陽性率為57.14%； 在對照組中，20例肝癌組織標(biāo)本中有18例AFP mRNA呈陽性表達，陽性率為90.00%，兩者間差異有顯著性意義（P＜0.05）。術(shù)后4周,希羅達治療組血清AFP濃度為（23.2±12.8） μg/L，明顯低于對照組的（39.6±24.3） μg/L（P＜0.05），而兩者術(shù)前血清AFP濃度差異無顯著性意義。
結(jié)論  希羅達能有效抑制人肝癌組織AFP mRNA的表達，并降低其表達產(chǎn)物血清AFP濃度。其在治療原發(fā)性肝癌的臨床應(yīng)用價值值得進一步研究。
