目的建立一種穩(wěn)定的急性壞死性胰腺炎模型。方法36只雄性ICR小鼠用查隨機數(shù)字表的方法隨機分為對照組(n=6)和實驗組(n=30)。實驗組予以雨蛙素腹腔注射(50 μg/kg),每小時注射1次,共7次,并分別于首次注射后9 h、18 h、24 h、48 h和72 h各處死6只; 對照組予以腹腔注射等量的生理鹽水,于注射后18 h處死。比較兩組動物的血淀粉酶值、胰腺重量及胰腺組織學(xué)評分分值。結(jié)果腹腔注射雨蛙素后9 h,小鼠血淀粉酶值升高,胰腺重量增加,18 h達峰值,24 h后逐漸下降。二者與胰腺病理損害程度相一致。18 h胰腺間質(zhì)炎性細胞浸潤和腺泡細胞壞死明顯,胰腺組織學(xué)評分值達到最大值(P<0.05)。結(jié)論大劑量雨蛙素腹腔注射能誘導(dǎo)ICR小鼠產(chǎn)生急性壞死性胰腺炎。該模型制備方法簡單,無創(chuàng)傷性,模型穩(wěn)定,重復(fù)性好。
