目的 觀察膽囊結(jié)石患者膽汁(CB)對人膽囊癌細胞系GBC-SD生長的影響,探討膽囊結(jié)石與膽囊癌發(fā)生與發(fā)展的關(guān)系。
方法 應(yīng)用四唑藍(MTT)比色法檢測20例CB和20例正常膽汁(NB)對GBC-SD細胞增殖的影響,應(yīng)用流式細胞儀測定細胞周期和凋亡。
結(jié)果 CB與NB比較,前者可明顯促進GBC-SD細胞增殖,用CB處理48 h后GBC-SD細胞增殖指數(shù)顯著上升(P<0.05)。CB組S期細胞比例為(49.26±8.07)%,明顯高于NB組的(25.54±6.57)%,P<0.05; CB組G0/G1期細胞比例為(40.59±9.12)%,明顯低于NB組的(60.64±13.42)%,P<0.05。CB組和NB組細胞凋亡率分別為(5.50±1.16)%和(3.91±1.76)%,P gt;0.05。
結(jié)論 CB具有潛在的促增殖活性,且膽囊結(jié)石與膽囊癌的發(fā)生與發(fā)展關(guān)系密切。
