目的 觀察氯化釓(GdCl3)對內(nèi)毒素刺激后的小鼠巨噬細胞來源的細胞系RAW264.7細胞Toll樣受體(TLRs)表達的影響。方法 RAW264.7細胞分為空白組、內(nèi)毒素處理組(LPS組)及GdCl3處理組(GdCl3組),采用流式細胞儀檢測TLR2/4蛋白表達情況; 用逆轉錄PCR(RT-PCR)法分析細胞中TLR2/4 mRNA表達的變化; 用ELISA檢測細胞培養(yǎng)上清液中TNF-α的水平。結果 與LPS組相比,不同濃度的GdCl3作用于RAW264.7細胞后,其TLR2/4蛋白和基因的表達以及TNF-α的表達水平均明顯下降,在觀察濃度范圍內(nèi)以2 000 μmol/L時最明顯,TLR2/4蛋白: 200 μmol/L時為(70.2±1.28)%/(66.7±2.59)%,400 μmol/L時為(64.9±1.43)%/(60.4±1.25)%,2 000 μmol/L時為(47.4±0.98)%/(32.1±0.74)%,其與 LPS組的(94.4±1.76)%/(95.7±0.87)%比較,P<0.01; TLR2/4 mRNA (A值): 200 μmol/L時為(76.42±2.76)/(101.72±3.14), 400 μmol/L時為(75.60±3.76)/(89.65±5.17),2 000 μmol/L時為(64.22±4.67)/(78.44±4.88),其與 LPS組的(127.64±3.25)/(119.82±5.59)比較,P<0.05, P<0.01; TNF-α: 200 μmol/L時為 (2 540±77) pg/ml, 400 μmol/L時為 (2 041±106) pg/ml,2 000 μmol/L時為(1 020±220) pg/ml ,其與 LPS組的(4 688±127) pg/ml比較,P<0.01。結論 GdCl3能明顯抑制內(nèi)毒素引起的RAW264.7細胞Toll樣受體的表達及相應炎癥因子的生成。
