【摘要】 目的 研究反義核酸技術(shù)封閉谷氨酰胺酶( GA) 基因?qū)β闶篌w內(nèi)胃癌細胞凋亡作用的影響。方法 將攜帶GA 反義基因的質(zhì)粒轉(zhuǎn)染胃癌細胞SGC27901 ,并將成功轉(zhuǎn)染的胃癌細胞接種于裸鼠皮下,獲得裸鼠胃癌移植瘤模型,通過TUNEL 技術(shù)檢測胃癌細胞凋亡的凋亡指數(shù); RT2PCR 技術(shù)檢測移植瘤細胞內(nèi)GA mRNA 的含量。結(jié)果 GA 反義基因質(zhì)粒載體成功轉(zhuǎn)染胃癌細胞后,腫瘤生長速度減慢,癌細胞凋亡增加,腫瘤細胞內(nèi)GA mRNA的含量減少。結(jié)論 GA 反義基因可抑制GA 基因的表達,明顯促進胃癌細胞凋亡。
