目的 在膽管結(jié)扎(BDL)模型中觀察神經(jīng)生長因子(NGF)及其高親和力受體酪氨酸激酶A(TrkA)在增殖的膽管周圍結(jié)締組織中的表達(dá)以及梗阻緩解后其表達(dá)的變化。
方法 46只SD雌性大鼠,隨機分為對照組( n =6)和BDL組( n =40),建立大鼠阻塞性黃疸模型,取肝十二指腸韌帶,采用免疫組化方法檢測NGF和TrkA在膽管結(jié)締組織中的表達(dá)和分布。同時左心室采血檢測血清總膽紅素和谷丙轉(zhuǎn)氨酶水平。
結(jié)果 BDL組膽總管結(jié)扎后,大鼠膽紅素水平于早期升高: 3 d時有明顯升高,持續(xù)到第14 d; 后期下降: 第21 d和28 d時膽紅素水平已基本降至正常。BDL組膽管壁直徑明顯增加,與結(jié)締組織細(xì)胞增殖分化相平行。NGF及TrkA受體在膽管結(jié)締組織細(xì)胞和炎性浸潤細(xì)胞胞膜和胞漿中表達(dá),其表達(dá)的變化與膽紅素水平的變化趨勢基本一致。
結(jié)論 NGF和 TrkA受 體對膽管周圍結(jié)締組織的增殖、分化有調(diào)節(jié)作用,可能參與了膽管瘢痕的形成過程,淤膽狀態(tài)的緩解在短期內(nèi)可能對膽管瘢痕形成無逆轉(zhuǎn)作用。
