目的:探討雌激素影響人精子頂體反應的可能機制。方法:應用異硫氰酸熒光素標記豌豆凝集素熒光染色法(FITC-PSA)分析精子頂體反應(AR)、以分光光度比色法測定頂體酶(ACE)活性。結果:17β-雌二醇(17β-E2)可促進精子發(fā)生AR,并增強精子頂體酶的活性;去除培養(yǎng)液中的Ca2+后,17β-E2不能誘導精子發(fā)生AR;PKC抑制劑能明顯降低17β-E2所誘導的AR;E2-BSA亦能夠促進精子發(fā)生AR,其作用與17β-E2無顯著差異。結論:雌激素對人精子頂體反應有一定的促進作用,增強精子頂體酶活性可能是其作用途徑之一,此過程涉及了胞外Ca2+、PKC及精子膜上的ER或雌激素結合位點的參與。
